It has been proposed that the inappropriate release of oxidants and proteolytic enzymes from activated neutrophils is responsible for joint damage observed in rheumatoid arthritis (RA). The present study examines the effect of increasing concentrations of synovial fluid [SF] (0-80%) on the oxidative response of neutrophils isolated from SF of RA patients and the blood of healthy subjects. To reflect better the real conditions of the inflamed joint we used in our experiments higher concentrations of SF than was previously reported. We found that increasing the concentrations of SF has a different effect on lucigenin-dependent and luminol-dependent CL response.
