abstract="The majority of drugs are metabolized by the liver, an organ responsible for detoxication. Each drug has its own pathway of biotransformation involving one or more cytosolic or membrane-bound enzymes. Most drug-induced liver injuries (DILI) result in hepatocyte necrosis however, some of them can comprise damage to the bile ducts, vascular endothelial cells or stellate cells. The mechanisms of DILI are divided into two fundamental groups, i.e., direct chemical reactions and idiosyncratic reactions. Some particular molecular mecha-nisms responsible for DILI have been explained, although there are many that are still unclear. In this review we discuss the issue of drug hepatotoxicity with special attention put on its mechanisms.",
