It has been shown to exert a potential for anti-cancer, anti-inflammatory and anti-fertility effects. Following effects of gossypol acetic acid (GA) in mice were determined in the present study: influence on the total number of lymphocytes, weight ratio and subsets of lymphocytes in the thymus, spleen and mesenteric lymph nodes, and humoral immune response to sheep erythrocytes (SRBC). Gossypol acetic acid was administered orally four times at 24 h intervals at three doses of 5.0, 25.0 and 125.0 mg/kg to SRBC-immunized mice (prior to or after priming), and non-immunized mice. It was found that GA decreased the total number of lymphocytes in thymus and mesenteric lymph nodes. Gossypol acetic acid dependent on the dosage decreased the percentage and absolute count of CD4+ thymocytes but increased the percentage and absolute count of CD8+ lymphocytes in spleen and lymph nodes. In SRBC-immunized mice, GA at the doses of 25.0 and 125.0 mg/kg administered prior to SRBC reduced the number of plaque forming cells (PFC) and the production of IgG. However, GA administered after priming decreased the production of IgM and IgG. In vitro, GA inhibited proliferation of J774.E murine macrophages in a dose- and time-dependent manner.
