(Pro)renin and angiotensin-converting enzyme 2 – new pathophysiological and therapeutic aspects of the renin-angiotensin system

In recent years it was proved that (pro)renin is not only a protease generating angiotensin-1-10 from angiotensinogen, but possesses its own receptors, which when activated lead to synthesis of cardiovascular and renal fibrotizing molecules bypassing angiotensin 2 generation. In addition a new angiotensin-converting enzyme 2 (ACE-2) was identified, which by degradation of angiotensin-1-10 or angiotension-1-8 respectively leads to formation of angiotensin-1-7. The latter, after binding with its specific mas receptor, initiates synthesis of peptides, which show biological effects quite opposite to those evoked by angiotensin-1-8. Finally ACE-2 proved to be a receptor for the coronavirus, which is the cause of SARS (severe acute respiratory syndrome). Thus therapy of cardiovascular, renal and some viral diseases, in whose pathogenesis the RAAS is involved, became enriched by new drugs.
