We present a brief historical introduction of tumour necrosis factor (TNF). Molecular structure and cellular origins are introduced. The TNF superfamily with its multiple members and variety of functions is summarized. Receptor activity and biological effects of TNF are discussed. Particular attention is drawn to TNF’s influence on neoplasms and inflammatory bowel diseases. Finally, anti-TNF therapies (including biological and small-molecule therapies) are described.
